The Veteran s Administration (VA) Office of Research and Development, Regional Procurement Office East in Pittsburgh, PA intends to negotiate on a sole source basis with Sophion Bioscience Inc., based in Woburn, MA on behalf of the VA West Haven Healthcare System. The requirement is for one (1) Automated electrophysiology instrument with specifications that meet or exceed those listed in the minimum specifications section. This instrument shall be delivered and installed at the Connecticut VA Medical Center for Neuroregeneration and Repair laboratory. Required system components: Sophion: SB1110 Qube 384 MkII SB3010 QVac SB3362 Temperature Control SB3350 Current Clamp SB3320 Rs Compensation for Qube Shipping and Insurance Minimum Specifications The vendor shall supply new, or refurbished, instrumentation that meets or exceeds the following specifications: A fully integrated, automated electrophysiology system for recording from 384 cells simultaneously. The instrument shall be able to
perform electrophysiological recordings in both voltage-clamp mode and current-clamp mode. Shall be fully compatible with an existing Sophion Qube 384 automated electrophysiology instrument currently installed in the laboratory. It shall utilize the same software and consumables and be fully interoperable to act as the backup for the current instrument in case of an interruption due to need for service. The proposed system shall provide an equal/better system then the Sophion advanced software solution in the field of automated patch clamp. The VA group have already gained deep understanding of the current Analyzer software and it is critical that this analysis can be continued using the same or completely compatible software. Shall have software that is traceable. This is required for proper documentation available in case a drug should go through approval process at the FDA. It is critical that the solution in the consumable can be completely exchanged. Open well consumables only allow to dilute compounds in the well and it is further impossible to wash out a compound completely. Sophion`s consumable include a microfluidic flow channel that allows to exchange the solution completely, so the proposed shall meet/exceed the current specifications. VA scientists work in multiple areas; therefore, having verified unattended operation is important. The system shall have the ability to run more than one plate without any operator intervention or hands on required. This requires not only automatic plate handling but further necessitates an on-board cell reservoir in which cells can be kept in media. The unit shall also include a centrifuge that automatically exchanges the media with the experimental buffer. Shall have individual electrode pairs built into each well allowing for a fresh set of electrodes for each new run and does not require the user to physically chloride electrodes regularly as in other systems. When working with voltage-gated sodium channels it is sometimes seen that double peaks are being recorded that arise from an artefact. It is critical that the software package includes an algorithm to detect double peaks and exclude this data. This would otherwise require a tremendous amount of time to go over all 384 wells. When testing compounds on voltage-gated sodium channels, it is important to also consider state dependent inhibition. The proposed system shall have the ability to determine the half inactivation potential (V1/2) for each cell and apply this value at a later point in the experiment. This leads to a lower scattering of the data and reduction of falsely characterized compounds. Must be able to add up to 24 different internal solutions in one experiment to accelerate the workflow. The requirement will take place at a VA facility Connecticut. The sole source is pursuant to FAR 6.302-1 Only One Responsible Source and No Other Supplies or Services Will Satisfy Agency Requirements. This notice of intent is not a request for competitive proposals, however; any responsible source who believes it is capable of meeting the requirement may submit a capability statement and supporting documentation to the contracting office no later than Monday, Nov 22, 2021, (10:00 AM, EST). A determination not to compete the proposed requirement based upon the responses to this notice is solely within the discretion of the Government.
