List of required specifications: Detection modes UV-Vis Absorbance Fluorescence intensity (must have top and bottom Fluorescence reading ability) Luminescence Detectors Each detection mode must have its own dedicated detector for increased sensitivity. Fluorescence detector: Dedicated PMT Absorbance detector: UV Silicon Photodiode Luminescence detector: Dedicated PMT Read method: Endpoint, kinetic, area scanning, absorbance spectral scanning Must have Well Scanning features that allow multiple read points per well (round, square, vertical or horizontal) Must incorporate internal auto spectral correction Must have fully automated Z focusing capability Light Source Dedicated Xenon flash lamp The system must have only one light source to minimize maintenance expenses UV-Vis Absorbance Wavelength range: 200 1000 nm, in nm increments Dynamic range: 0 to 4.0 OD Must have full 200-1000nm spectral scan in 1nm steps performed in under 5 sec Fluorescence Sensitivity: 2 pM fluorescein Excitation filters: 230 - 900nm & Emission filters: 280 - 900nm; bandwidth depends on filter used Optical mirrors: 50%, 410, 430, 458, 510, 560, 593, 625, 660 nm - must be user selectable Filter slides must be independently selectable to allow easy filter swapping without tools Dichroic mirrors must be selectable via software without manual user intervention Luminescence Wavelength range: 370-700nm Dynamic range: >9 orders of magnitude Sensitivity: 12 amol/well ATP OD1, OD2, OD3 attenuation filters Must have OD attenuation filters (3) enabling a dynamic range of 9 orders of magnitude for optimizing the widest range of luminescent signal intensities Must use dedicated fibers for luminescence in each plate format minimizing cross-talk between wells. Measurement Speed: 22sec, 384-well plate Plate Formats: 6- to 384-well, compatible with cuvettes; Roboflasks; cell chips; NanoQuant plate Shaking: Linear, Orbital, and Double-Orbital with variable amplitudes and frequencies Movement, speed and amplitude must be user selectable. Temperature Incubation: Ambient +3°C to 42°C optimized to eliminate edge well effect Must have optional module that allows maintaining ambient or sub-ambient temperatures in measurement chamber. Software: Must be able to select individual non-continuous wells (RAW data acquisitions sent directly to MS-Excel© including measurement parameters) Must provide workflows via condition strips, which will allow for increased workflow automation and controls. Must have a plate in and out button independent of software connection Connectivity: UBS 2.0 port Upgrade potential: Must be fully modular and able to be upgraded with modules for: Evaporation Protection including: The capability to lift the plate out when lifting the lid Small and large humidity cassettes Automated Cell Imaging including: Bright-field with 4x objective Laser based autofocus Cell counting and confluence functionality Contracting POC is Scott James at scott.james@va.gov Responses are to be submitted to scott.james@va.gov NLT 11:59am PST on 2/8/2022
