DESCRIPTION: The National Institutes of Health - NICHD intends to procure the services of a contractor to provide a EnVision Xcite Plate Reader system with software and related components as stated below (SOW).   Envision 2105 Xcite configuration will contain a single PMT Unit with Fluorescence Intensity (top and bottom), Absorbance, Standard Luminescence, Fluorescence Polarization, Time-Resolved Fluorescence/FRET. The configured Envision 2105 Multimodal Plate Reader will also include EnVision Manager Software, Win-10 Desktop Computer and 17'' TFT Flat Screen Monitor. The  configuration additionally includes our Ultra- Sensitive Luminescence and a Temperature Control Module Optics included in the requested configuration include those for GFP and mCherry assay reads, as well as filters to read absorbance at 420 nm, 550 nm, 595 nm and 600 nm. Additional Envision Optics can be quoted that include various Mirror Modules, Excitation filters, and Emission filters that will allow your
Envision 2105 to operate in filter-based mode for the utmost flexibility and sensitivity. Included in your purchase of the Envision Multimodal Plate Reader will also include installation, training, local Sales and Application Support as well as a one year all-inclusive warranty. All servicing and support of the Envision 2105 will be performed on-site to minimize any experimental delays.Unique Features of the PerkinElmer Envision Multi-Label Plate Reader:  The Reader must include or be field upgradeable with the following Detection Technologies: o AlphaScreen/AlphaLISA TechnologyUltra-Sensitive LuminescenceStandard LuminescenceFluorescence Intensity Top and BottomTime-Resolved FluorescenceTime-Resolved FRETAbsorbance (UV & Visible)Fluorescence Polarization Filter-based and Quad Monochromator-based Absorbance and Fluorescence Intensity tunable in 0.1nm increments Temperature Control from 2 degrees above ambient to 55 degrees C.Alpha Technology must use Laser Excitation at the standard 680nm Laser. This yields a significant increase in sensitivity and speed of detection and may be critical for assays which require lowered limit of detection samples.▪ Alpha Technology: System must be capable of Alpha detection at elevated temperaturesfrom 2 degrees C above ambient to 55 degrees C.▪ Alpha Technology: Two separate options are available; Alpha HTS "Turbo" Option for high throughput and Alpha Standard Option for low-medium throughput. They both function with single emission reading and are therefore suitable for both single and dual detector models. The typical measurement times when measuring Alpha on a 96-well plate are 30 seconds / plate with Alpha HTS Option and 68 seconds / plate with Alpha Standard Option. Multiplexing is only possible on the Standard Option. Multiplexing can be performed on the dual detector model in parallel, on the single detector subsequently, resulting in ~2 times slower reading time.▪ The Reader must include or be field upgradeable with the following Additional Hardware: ▪ Time-resolved Fluorescence and TR-FRET Laser with focused Excitation at 337nm▪ Quad Monochromator Setup to combine the sensitivity of a filter-based reader with the wavelength scanning and versatility of a monochromator-based reader. For Fluorescence Intensity, the 2105 Model will contain 2 Excitation Monochromators and 2 Emission Monochromators.▪ Ultra-sensitive Luminescence with dedicated PMT▪ Robotic Microplate Stackers with choice of 20 or 50-plate Magazines Dedicated PMT Detector for Ultra-Sensitive Luminescence detection with the PMT situated in close proximity to the sample microplate. This yields a significant increase in sensitivity and speed of detection, and may be critical for assays which require detection of low-expressing samples. o Ultra-Sensitive Luminescence is 30X more sensitive than standard luminometers▪ The System must be upgradeable with dual PMT Detectors for Fluorescence Detection/Time-resolved Fluorescence/TR-FRET, enabling fast detection of Dual-emission assays such as FRET and BRET.▪ Optimization Wizard to automatically determine optimal settings for the following parameters. This will greatly improve the performance of the reader and accelerate assay development: o Microplate Dimensionso PMT Detector Sensitivityo Excitation Lamp Powero Crosstalk Correction for Luminescence Detectiono Minimal number of flashes for targeted Z-Prime valueo Flat-field Correctiono Read Height▪ Use of Application-specific Dichroic Mirror Modules increases Sensitivity of TRF and TR-FRET assays such as DELFIA and LANCE Modules.▪ Up to four optimized Dichroic Mirror modules can be installed at once.▪ Up to eight (8) excitation/absorbance filters and eight (8) emission filters can be installed at one time.
